In the present study, two experiments were carried out. In the first experiment, insulin coated using Nano-polymer in order to calculate its duodenum absorption in sheeps. Scine the parenteral administration is the only route of insulin delivery, alternative route of administration (oral, nasal, rectal, pulmonary and ocular) have been extensively investigated among them, the oral route seems to be the most convenient and physiological because insulin undergoes a first hepatic glucose output. However, insulin is strongly degraded by proteolytic enzyme in the gastrointestinal tract. In addition, this peptide is very poorly absorbed after oral administration. In order to protect it from biodegradation and to improve its intestinal absorption, insulin has been associated to antiproteases, hydrogels, or combined with absorption enhancers such as cyclodextrins, bile salts and surfactants. Insulin was also encapsulated in polymeric biodegradable nanocapsules, nanoshperes or microparticles associated or not to surfactants or antiproteasas. In this study, nano particle were manufactured according to the double emulsion technique mainly used for peptides and proteins. Then many attempts have been conducted to make new formulation for orally insulin and decrease the consequence of insulin injection daily. To improve the stability of insulin against gastrointestinal enzymes, it was coated by using Edrugait polymer. Then Nanopolymer coating insulin was tested in rabbit as animal model. This Nano polymer decreased plasma glucose level. After testing, Nano-polymer on rabbit, they were delivered orally to 15 sheeps, for 5-day period. Blood samples were collected three times. The concentration of cortisol, estrogen, insulin and IGF-I were analyzed. The results showed that concentration of cortisol were significantly in higher level and progesterone in lower value. Cortisol was 2.7 µg/dl and progestron was 2.5 ng/ml. In the second experiment IGF-I was coated as Insulin did. As above follow Nano-polymer of IGF-I decreased the level of plasma glucose in rabbit. In order to accessibility to duodenum of sheeps, four sheeps were surgery and studied for 5-day period. Blood samples were collected on the first, third and fifth day of period. The concentration of progesterone and IGF-I were significantly in higher level. Progestron was 6.2 ng/ml and IGF_I was 69.5 ng/ml. Then we delivered Nanopolymer of IGF-I orally to 15 sheeps in a 5-day period. The concentration of cortisol, estrogen, insulin and IGF-I were measured. The concentration of cortisol increased significantly and progesteron and cortisol decreased. Cortisol was 2.8 µg/dl, Stradiol was 0.28 ng/ml and progestron was 2.9 ng/ml.
