In this thesis sol–gel process is used for manufacture of polydimethylsiloxaneadsorbent on steel wire. After that tramadol drug was extrated by the solid phase microextraction, and analyzed by Ion mobility spectrometry. Aninjection chamber wasconstructed for connection of SPME techniqueto ion mobility spectrometry. In this design, sample input, gas inletand element location is on one piece. In this project the dead volume of injection chamber is low so that the sample leaves the injection chamber and quickly enterd to the ion mobility spectrometer. Therefore, the peakbroading can be minimized. Extraction parameters such assalt concentration, temperature, equilibrium and extraction time were studied. The best extraction efficiency was observeat saturated salt concentration, 15 mMof NaOH,Temperaturewas , equilibrium time was 2 min and the extraction time was 10 min. Calibration curves were plotted in water, urineand plasma sampels. For water and urine samples linear daynamic range was obtained from 2-100?g/L and r 2 was0.9982 and 0.9976 for water and urine sampels, respectively.Linear daynamic range was obtained from 20 to 700 ?g/L andr 2 equal 0.9980for plasma sample.The limit ofdetection was obtained 1?g/L and 9 ?g/L in analisis of urine and plasma samples, respectively.Relative standard deviation in urine and plasma samples was8%and 10%, respectively. For the ability of the method in analysis of real samples, urine and plasma samples of volunteer analyzed by the above method after toking tramadol.
