Aleurone layer is a thin layer on grain seeds which has a key role in germination of the seed. The possibility of separating aleurone layer of barley seed from other layers, and also cultivating it on an appropriate buffer, attracted researchers to use it as a model system to study plant signals in molecular level. By far, using this system, the amount of synthesis and the effects of different protein enzymes in growth and germination of the seed have been studied. Among these is a study on the effect of gibberelic acid (GA) hormone on production of Alpha amylase enzyme, as the most important enzyme in starch endosperm hydrolyses. But there is no reference to the effects of ethylene hormone on the synthesis and secretion of alpha amylase and limit dextrinase (LD), both have been created in aleurone layer and in response to GA hormone. Present paper tries to study the effects of ethylene hormone on synthesis and secretion of induced proteins with gibberellic acid hormone, from aleurone layer. To that aim, aleurone layer has been incubated for 24 hours in 5m Molar GA hormone. Then, without changing the buffer, in the presence of GA hormone ethephon, in different density (0/1, 1, 10, 100, 250, 500, 1000, 2000, 5000 and 10000 ppm ), has been added. After extracting the protein, the solution from aleurone layer has been run on SDS-PAGE gel. Also, the cultivation buffer related to each treatment has been collected and densified with deposition and run on SDS-PAGE gel. Results show that with increase in the density of ethephon up to 10000 ppm, the total amount of the protein remained in the aleurone layer decreases, and on the other hand the amount of proteins extracted from aleurone layer has significantly increased. To study the quantitative changes, the amount of solvable proteins have been densified by Brad Ford method. In order to determine the effect of ethephon hormone on synthesis and secretion of alpha amylase enzyme, the Western Blot Test has been used with the help of related anybodies. The experiment shows that with the increase in the density of ethephon, the amount of secretion of both enzymes increased. Regarding the effect of ethephon on the increase in the activity of xylanase, which has been prove in previous works, the secretion of proteins by ethephon can be considered as the effect of ethylene hormone on the structure of cell wall in aleurone layer. The inductive effect of ethephon on protein secretion regarding the LD enzyme has also been reported, in such a way that despite there is no such protein with aleurone layer treated with GA after 24h, and the secretion has been delayed in treatment with GA up to 72h, in the presence of ethephon it has also been induced in the extraction buffer.
