Potato ( solanum tuberosum L.) is a heterozygous and autotetrapliod plant and one of the tuber crops that very important for feeding the people over the world. The potato produces more protein and calories per unit area after wheat and rice. As will as it is considered the fourth major crop after wheat, rise and maize economically. Since the cloning of potato increases viral disease spread especially viruses “Potato leaf roll virus ( PLRV ) and potato virus Y ( PVY )” and those viral diseases constitute a major constraint to high yield and high quality production of potato. In connection with the point, producing virus free plants through micropropagation technology combinined with elemination techniques including chemotherapy, cryotherapy or electrotherapy for efficient virus eradication is emphasized. In the present investigation attempts were made to evaluate the effects of different levels of NAA ( 0, 0.1, 0.3 and 0.5 mgl -1 ) and BAP (0, 0.2 and 0.4 mgl -1 ) on sprout tip (1, 2 and 3 mm) culture of two potato cultivars “ Chandramokh and Marfona ”, and to eliminate PLRV and PVY viruses from infected vitroplants by chemotherapy and cryotherapy. Results of the former indicated that the maximum plantlet length (151.66 mm and 146.66 mm) was observed in “ Chandramokh” cultivar in the treatment NAA and BAP (0.3, 0.4 and 0.5, 0.2 mgl -1 with explants size of 2 mm, respectivily. and in Marfona cultivar the highest plantlet length was obtained in 0.5 mgl -1 NAA and 0 mgl -1 BAP with the sprout tip size of 3 mm. The highest number of leaves and nodes per plantlet were recorded in the cultivars “ Chandramokh” and “ Marfona” at NAA and BAP (0.1, 0.0 and 0.3, 0.0 mgl -1 ) with the explant size 2 and 3 mm in length, reectivily. The highest number of roots were obtained in the cultivar “ Chandramokh” at NAA and BAP (0.5, 0.2; 0.5, 0.0 and 0.1, 0.0 mgl -1 ), and the largest root length was observed at 0.1 mgl -1 of NAA and BAP at 0.0 mgl -1 with sprout tip 2 mm in size, in the cultivar “Marfona”, the highest number of roots and root lengths was observed in the treatment NAA and BAP at 0.3, 0.0 and 0.5, 0.0 mgl -1 with the sprout tip 3 mm in length. In the latter, the segments of sprout tip (3.0 and 5.0 mm in size) from infected mother plant (indexed by ELISA and RT-PCR ) were culture on MS medium suplimented with BAP (0.0 mgl -1 ) and NAA (0.1 mgl -1 ) containing Ribovirin ( RBV ) and Azacytidine ( AZA ) each at 0.0, 20, 40 and 60 mgl -1 .
