Some of the small molecules such as Purmorphamine play an important role in the Sonic Hedgehog (SHH) signaling pathway. Purmorphamine is an agonist of SHH, which plays a critical role in the regulation activity of receptors, transcription factors and also can be used to confirm the Sonic Hedgehog pathway. In this study, effect of Purmorphamine on oocyte maturation and epigenetic modification of in vitro matured sheep oocytes were considered. Ovaries were obtained from local abattoir and cumulus-oocyte complexes cultured in the medium containing various concentrations of purmorphamine (250 and 500ng) plus a control without purmorphaminel. After 24h maturation, oocytes striped from cumulus cells and stained with Hoechst and cell tracker blue for assessment of nuclear and cytoplasmic maturation respectively, fluorescent microscope was used for photograph, taken digital images analyzed by Image-J software. For fertilization, 22 h after maturing the oocytes, they were transferred to fertilization medium and co-incubated with sperm for 18 h. Two cell embryos were used for RNA extraction and total RNA for Real Time PCR. The records were analyzed by REST software. Developmental competence oocyte examined using artificial activation, cleavage and blastocyst rate measured on D3 and D8 post activation respectively. The results showed that 250ng of Purmorphamine induced nuclear maturation and about 81 percent of oocyte showed meiosis II stage, but not significantly different from the control group (p 0.05). Based on statistical analysis 500 ng purmorphamine reduced expression PTC but SMO and GLI significantly increase (p 0.05). Furthermore, it was observed that the expression of Histone deacetylase 1-3 was reduced. Acetylation of histone H4 lysines 12 was significantly higher in 500 ng Purmorphamine. Besides, the results showed that in the presence of purmorphamine, embryo development was significantly increased. In conclusion, purmorphamine induces cytoplasmic maturation of oocyte and can be used to confirm the Sonic Hedgehog pathway.
