Pathogenesis Related proteins are compounds is produced in response to pathogens or Environmental stress. These proteins can be classified into seven teen group by amino acid sequences, serological differences and Enzymatic kinetic among these classes 1,3-?-Glucanase and Chitinase are most studied be cause of 1,3-?-Glucanase by hydrolyzing1,3-?-Glucan bond and Chitinase by hydrolyzing 1,4-N acetyl amin bond can directly hydrolize fungal cell wall and inhibit fungal growth. The aim of this research was Isolation, over-expression analysis of physiological activity of ?-1,3- glucanase from Medicago truncatula. Until now numerous kind of 1,3-?-Glucanase are Identified in various plants and thein fungicide effect were tested. Watson et al reported 304 protein with peptide mass finger printing technique in medicago truncatula among them a 1,3-?-Glucanase was registered with BF 650622 Id number. But until now It,s Biological activity had ot been studied. In this case by Medicago sativa and Pisum sativum 1,3-?-Glucanase Gene alignment on Medicago truncatula Genome three open reading frame with 50% similarity by having start and stop codon was identified and isolated. Then these fragments was cloned in PET21c vector and expressed in E.coli BL21. SDS PAGE technique reveal that proteins were produce as an Inclusion bodies. We also test their fungicide effect after folding them to achieve their natural structure on Alternaria alternaria and Fusarium graminearum. The result were significant ( P 0.05 ) by SAS system analysi
